In particular, the path for sure does not realy start with "/Desktop"...

You might want to try

   dir(file.path(Sys.getenv("USERPROFILE"), "Desktop/work")) and correct the entry to

   read.table(file.path(Sys.getenv("USERPROFILE"), "Desktop/work/128_L.YourExtension"))
